Dr. David Nemazee is a Professor in the Department of Immunology and Microbiology at Scripps Research and a recipient of the 2025 Crafoord Prize in Polyarthritis. Awarded by the Royal Swedish Academy of Sciences, this prestigious honor recognizes his discovery of the fundamental molecular mechanisms governing B-cell tolerance.

Dr. Nemazee is globally celebrated for his pioneering identification of receptor editing, a landmark finding that overturned long-standing immunological dogmas by showing how self-reactive B cells can secondary-recombine and reset their specificity to avoid attacking host tissues. This work provides the essential mechanistic foundation for our modern understanding of systemic autoimmune diseases like rheumatoid arthritis and lupus.

Expanding his focus on immune homeostasis into the realm of innate immunity, Dr. Nemazee’s laboratory recently redefined endosomal regulation through the discovery of PLD4 (and its paralog PLD3) as a critical endolysosomal 5' exonuclease. By functioning as a molecular scavenger that degrades single-stranded DNA before it can aberrantly activate TLR9 signaling, PLD4 serves as a vital gatekeeper against nucleic-acid-driven autoinflammation. Beyond these foundational contributions to adaptive and innate tolerance, the Nemazee laboratory remains a leader in translational immunology, utilizing advanced single-cell sequencing to design germline-targeting strategies for next-generation vaccines against highly mutable pathogens such as HIV-1 and influenza.
